Abstract

Gallium-67 tumor scanning is widely employed in the clinical detection and staging of a broad range of solid tumors and malignant lymphomas. As a basis for understanding the molecular mechanism of gallium localization in tumor cells, the aqueous chemistry of gallium(III) was investigated by nuclear magnetic resonance (NMR) spectroscopy. Specifically, these studies include: characterization under various experimental conditions (pH, concentration, temperature etc.) of gallium citrate complexes, of gallium phosphate complexes, and of gallium complexes with EDTA (ethylenediaminetetraacetate), NTA (nitrilotriacetic acid) and lactate etc. The results of these studies demonstrate that all the agents which inhibit in vitro gallium-67 uptake by L1210 leukemic cell also form complexes with the metal under various experimental conditions. The extent of inhibition of gallium-67 uptake correlates semiquantitatively with the relative stability of these complexes. This suggests that the ability of these agents to prevent localization of gallium-67 in tumor cells results from formation of complexes with the metal, which are either impermeable to the tumor cell membrane or are more stable than intracellular gallium complexes.
